تسریع روند ترمیم استخوان از طریق تزریق مستقیم محیط‌های کشت تمایز دهنده ‌استخوانی و معمولی همراه با هیدروکسی آپاتیت در نقیصه استخوانی رادیوس در موش صحرایی

چکیده

پیشینه: ترمیم نقص‌های استخوانی بحرانی همچنان یکی از چالش‌های اصلی جراحان ارتوپدی است. مطالعات متعددی به بررسی روش‌های بهبود ترمیم استخوان پرداخته‌اند. هدف: هدف این مطالعه، ارزیابی توانایی ترمیمی محیط‌های کشت سلولی تمایز دهنده‌ استخوانی و نگهدارنده در صورت تزریق مستقیم به محل نقص، به تنهایی یا همراه با گرانول‌های هیدروکسی آپاتیت (HA) بود. روش‌ کار: هفتاد موش نر ویستار در هفت گروه تقسیم‌بندی شدند: نقیصه خالی (10 عدد، کنترل منفی)، اتوگرافت (10 عدد، کنترل مثبت)، HA (10 عدد)، محیط کشت استخوانی (OM) (10 عدد)، محیط‌کشت معمولی (MM) (10 عدد)، هیدروکسی آپاتیت و محیط کشت معمولی (MM+HA) (10 عدد)، و هیدروکسی آپاتیت و محیط‌ کشت استخوانی (OM+HA) (10عدد). یک نقیصه‌ ۵ میلی‌متری در دیافیز استخوان رادیوس به عنوان نقص بحرانی ایجاد شد. محیط‌های کشت در روز چهارم پس از جراحی به محل نقیصه تزریق شدند. ارزیابی‌های رادیولوژیک در روزهای ۱۴، ۲۸، ۴۲ و ۵۶ پس از عمل انجام شد. بررسی‌های آسیب‌شناسی بافتی در روزهای ۲۸ و ۵۶ صورت گرفت. نتایج: محیط‌های OM و MM، چه به تنهایی و چه همراه با HA، توانایی قابل توجهی در ترمیم استخوان نشان دادند. از نظر رادیولوژیک، تمامی گروه‌های درمانی نسبت به کنترل منفی ترمیم بهتری داشتند. بازسازی استخوان در گروه OM در روزهای ۴۲ و ۵۶ با گروه اتوگرافت قابل مقایسه بود. از نظر آسیب‌شناسی بافتی، تمامی گروه‌ها به‌جز گروه HA در روزهای ۲۸ و ۵۶ روند ترمیم پیشرفته‌ای را نشان دادند. به‌ویژه، ترمیم استخوان در گروه‌های HA+OM و HA+MM در روز ۲۸، و در گروه‌های OM و MM در هر دو روز ۲۸ و ۵۶ مشابه گروه اتوگرافت بود. نتیجه‌گیری: محیط‌های کشت تمایز دهنده‌ استخوانی و معمولی دارای ویژگی‌های امیدوارکننده‌ای در بازسازی استخوان هستند و در صورت توسعه سامانه‌های مناسب برای انتقال و حمایت، می‌توانند به عنوان درمان‌های مؤثر برای نقص‌های استخوانی مورد استفاده قرار گیرند.
