اثربخشی انوکساسین و لووفلوکساسین علیه رده سلولی آلوده به شیزونت تیلریا آنولاتا

چکیده

پیشینه: تیلریوز یکی از بیماری‌های منتقله از کنه است که به طور قابل توجهی صنایع دامداری را در کشورهای در حال توسعه مختل می‌کند. بوپارواکون تنها گزینه موجود برای مدیریت تیلریوز است. از سوی دیگر، استفاده از دارو یا تکامل ارگانیسم منجر به بروز مقاومت در طول بیست سال گذشته شده است. درمان تیلریوز نیاز به رویکردهای درمانی جایگزین دارد. هدف: این کار به بررسی اثربخشی برون‌تنی ترکیبات انوکساسین و لووفلوکساسین علیه کشت‌های تیلریا آنولاتا با هدف قرار دادن مسیر توپوایزومراز II ارگانیسم می‌پردازد. روش‌ کار: رده سلولی آلوده به شیزونت از خون گاو آلوده تهیه شد. اثرات مهاری هر دو دارو بر روی تیلریا آنولاتا به همراه سمیت سلولی و فعالیت همولیتیک آن‌ها ارزیابی شد. نتایج: انوکساسین و لووفلوکساسین به ترتیب در غلظت‌های ۱۷۱۷۲۳ میکرومولار و ۱۰۰۸۴ میکرومولار به طور مؤثر 50% از عفونت را مهار کردند. لووفلوکساسین در طول آزمایش نتایج دلگرم کننده‌ای نشان داد. برای انوکساسین و لووفلوکساسین، اثرات سیتوتوکسیک 50% نیز به ترتیب در غلظت‌های ۱۰۵۲۹ میکرومولار و ۱۰۱۱۶ میکرومولار اندازه‌گیری شد. انوکساسین با مقدار HC50 معادل ۲۶۴۹۰۳۱ میکرومولار در مقایسه با لووفلوکساسین با مقدار ۱۹۸۲۸ میکرومولار، بر روی سلول‌های خونی گاو ایمن بود. نتیجه‌گیری: لووفلوکساسین در مقایسه با انوکساسین نتایج رضایت‌بخشی نشان داد. مطالعات بیشتر به همراه سایر پارامترها برای ارزیابی و سنجش فعالیت مهاری انوکساسین و لووفلوکساسین مورد نیاز است.
