شناسایی مولکولی سالمونلا جداشده از ماکیان تجاری

چکیده

پیشینه: سالمونلا در ماکیان خصوصا سالمونلاهای متحرک، زنجیره غذایی از مزرعه تا سفره را ناایمن کرده است و به عنوان یک تهدید جهانی مهم برای سلامت عمومی در نظر گرفته می‌شود. هدف: مطالعه حاضر به منظور شناسایی مولکولی سالمونلا در ماکیان تجاری با استفاده از PCR انجام شد. روش کار: این مطالعه به مدت هشت ماه از تیرماه تا بهمن ماه 1398 انجام شد و در مجموع از 26 مرغداری شامل 15 مزرعه گوشتی و 11 مزرعه تخم‌گذار به صورت انفرادی بازدید شد. نمونه‌های ادغام شده مدفوع از سوله‌ها تهیه شدند. در مجموع 189 مورد کالبد گشایی و از نظر ضایعات گوارشی مورد بررسی قرار گرفتند. جداسازی و شناسایی ارگانیسم با استفاده از روش کشت میکروب انجام شد. خصوصیات مولکولی با روش PCR و با هدف قرار دادن ژن‌های invA و ent انجام شد. نتایج: با استفاده از روش سنتی استاندارد کشت، شیوع سالمونلوز در مزارع گوشتی و تخم‌گذار به ترتیب 20% و 5/45% ثبت شد. از مجموع 189 مورد پرنده کالبد گشایی شده، سالمونلوز در 58/1% طیور مرده ثبت شد. شناسایی مولکولی جدایه‌های سالمونلا با استفاده از PCR هدف گیرنده ژن invA، در 33/13% مزارع گوشتی و 3/36% زارع تخم‌گذار تایید شد. شناسایی سالمونلا انتریتیدیس با استفاده از PCR هدف گیرنده ژن ent انجام شد که به وسیله آن 11/11% مثبت تایید شدند. نتیجه‌گیری: این مطالعه که بر شیوع سالمونلا تمرکز داشت، اهمیت قابلیت انتقال این باکتری را به انسان در مزارع تجاری پرورش ماکیان نشان داد است که متعاقبا می‌توانند وارد زنجیره غذایی انسان شده و اثرات مضری بر سلامت داشته باشند.
