چکیده

پیشینه: منهیمیا همولیتیکا در درجه اول سبب پنومونی در نشخوارکنندگان اهلی می‌شود که منجر به واگیری و مرگ و میر در سرتاسر جهان می‌شود. اخیرا، لیپوپروتئین‌های غشاء خارجی به عنوان اهدافی برای القاء ایمنی محافظت کننده بر ضد عفونت پاستورلا پدیدار شده‌اند. هدف: هدف از مطالعه حاضر، ارزیابی لیپوپروتئین غشاء خارجی نوترکیب (PlpE) E از جدایه منهیمیا همولیتیکا گوسفندی، به عنوان یک کاندید واکسن می‌باشد. روش کار: PlpE نوترکیب با استفاده از وکتور بیانی pET26 (b) در باکتری اشریشیا کلی ساخته شد. PlpE نوترکیب بیان شده تخلیص و زیر پوستی به موش تزریق شد. اثر محافظت کنندگی واکسن از طریق چالش داخل صفاقی موش مورد ارزیابی قرار گرفت. نتایج: پاسخ‌های آنتی بادی ضد PlpE به طور معنی‌دار در موش‌های ایمن شده در مقایسه با گروه کنترل افزایش یافت که چالش داخل صفاقی با سویه حاد منهیمیا همولیتیکا محافظت کنندگی موثری در موش‌های ایمن شده فراهم کرد. نتیجه‌گیری: PlpE نوترکیب از جدایه منهیمیا همولیتیکا گوسفندی پتانسیل آن را داشت که به عنوان یک کاندید واکسن ضد عفونت منهیمیا همولیتیکا در گله‌های گوسفند استفاده شود.
