اثرات سیترال بر وضعیت آنتی اکسیدانی سرم و بیان ژن‌های کبدی پاراکسوناز 1 و نیتریک اکسید سنتاز در یک مدل موش صحرایی دیابتی ملیتوس القایی با استرپتوزوتوسین

چکیده

پیشینه: سیترال (C10H16O)، ماده اصلی سیمبوپوگون سیتراتوس (روغن علف لیمو) است و می‌تواند عوارض جانبی استرس اکسیداتیو را کاهش دهد. دیابت ناشی از کمبود انسولین باعث استرس اکسیداتیو در کبد می‌شود. هدف: اهداف این مطالعه بررسی اثرات بهبودی سیترال بر پارامترهای اکسیداتیو انتخاب شده و بیان ژن پاراکسوناز 1 (PON1نیتریک اکسید سنتاز اندوتلیالی (eNOS) در یک مدل موش صحرایی دیابتی ملیتوس القا شده با استرپتوزوتوسین (STZ) بود. روش کار: چهل موش صحرایی به طور تصادفی به چهار گروه تقسیم شدند: کنترل (C)، کنترل سیترال (CC)، و دو گروه دیابتی ناشی از STZ (دیابتی (D) و سیترال دیابتی (CD)). پس از تایید دیابت (روز 7)، درمان از طریق گاواژ با سیترال (300 میلی گرم در کیلوگرم وزن بدن (BW)) در گروه‌های CD و CC آغاز شد و به مدت 2 هفته ادامه یافت. نتایج: در روز 21 مطالعه، به دنبال درمان با سیترال به مدت 14 روز، سطح سرمی ظرفیت آنتی اکسیدانی تام (TAC) و PON1 در گروه CD به طور معنی‌داری در مقایسه با گروه D افزایش یافت (P<0.05). در حالی که درمان با سیترال، سبب کاهش معنی‌داری در MDA، و eNOS در گروه CD نسبت به گروه D شد (P<0.001). میزان بیان ژن PON1 کبد در گروه CD نسبت به گروه D به طور قابل توجهی افزایش یافته بود (P<0.001)؛ در حالی که بر عکس آن برای بیان ژن کبدی eNOS مشاهده شد. با این حال، بین گروه‌های C و CC از لحاظ تمامی شاخص‌های مورد بررسی هیچ اختلاف معنی‌داری یافت نشد (P>0.05). نتیجه‌گیری: این مطالعه اثرات مثبت سیترال را بر وضعیت آنتی اکسیداتیو سرم و بیان ژن کبدی PON1 و eNOS در موش صحرایی دیابتی نشان داد.
