اثر سیستئامین در طول بلوغ آزمایشگاهی بر زنده مانی و توانایی میوز تخمک‌های منجمد شده بوفالو

چکیده

اهداف مطالعه حاضر بررسی اثرات سیستئامین به عنوان یک آنتی اکسیدان بر میزان بلوغ تخمک‌های بوفالو در محیط آزمایشگاه (آزمایش 1)، و زنده مانی و وضعیت هسته‌ای آن‌ها متعاقب انجماد شیشه‌ای (آزمایش 2) بودند. تخمک‌های نابالغ با سلول‌های متراکم کومولوس تهیه شده از تخمدان‌های حیوانات کشتار شده جمع‌آوری شدند و سپس در محیط کشت بلوغ بدون سیستئامین (کنترل) یا μM 50 سیستئامین (درمان شده) کشت داده شدند. تخمک‌ها در محلول انجماد شیشه‌ای 1 (VS1): M 5/1 اتیلن گلیکول (EG) + M 5/1 دی متیل سولفوکسید (DMSO) به مدت 45 ثانیه منجمد شدند (مرحله اول). بعد از این در معرض قرارگیری اولیه، تخمک‌ها به VS2: 3 M EG + 3 M DMSO در یک محیط نگهدارنده به مدت 25 ثانیه منتقل شدند (مرحله 2). بعد از ذوب شدن، تخمک‌ها از لحاظ مورفولوژی بررسی شدند و سپس به مدت 2 ساعت دیگر در محیط‌های کشت بلوغ کنترل یا مکمل سازی شده با سیستئامین کشت داده شدند. تخمک‌ها از لحاظ ظاهری مورد ارزیابی قرار گرفتند و با تریپان بلو برای بررسی زنده مانی رنگ آمیزی شدند. میزان بلوغ تخمک‌ها برای محیط کشت بلوغ آزمایشگاهی با سیستئامین در مقایسه با کنترل‌ها بیشتر بود (P<0.05). اختلاف معنی‌داری از لحاظ مورفولوژی، بقا و میزان بلوغ بین دو گروه انجمادی (گروه‌های درمان شده با سیستئامین و درمان نشده) وجود نداشت، اما مورفولوژی، بقا و درصدهای تخمک‌های متافاز II در هر دو گروه تخمک‌های منجمد شده در مقایسه با کنترل‌های مربوط به خود کمتر بودند. در نتیجه، افزودن سیستئامین به محیط کشت بلوغ، بلوغ هسته‌ای تخمک‌های بوفالو را بهبود بخشید، اما اثر مثبتی بر مقاومت به سرما در طول انجماد شیشه‌ای نداشت.
