تنوع ژنتیکی سویه‌های کلستریدیوم پرفرینجنس جدا شده از منابع غذایی و انسانی

چکیده

کلستریدیوم پرفرینجنس، یک بیماری‌زای مهم بوده که در حیوانات اهلی، بیماری‌های گوارشی و در انسان مسمومیت غذایی ایجاد می‌کند. اسپورها طی فرآیند پخت زنده مانده و نقش مهمی در شروع احتمالی بیماری ایفا می‌کنند. در این مطالعه RAPD-PCR و REP-PCR برای بررسی تنوع ژنتیکی 49 ایزوله کلستریدیوم پرفرینجنس تیپ A از سه منشا مختلف استفاده شدند. نتایج RAPD-PCR، بیشترین تنوع را در بین ایزوله‌های مرغی نشان داد در حالی که ایزوله‌های انسانی کمترین تنوع را نشان دادند. آنالیز گروه‌بندی به دست آمده از RAPD-PCR و بر اساس فاصله‌های ژنتیکی، 49 ایزوله را به 5 گروه اصلی A)، B، C، D و (E تقسیم نمود. گروه‌های A و C از ایزوله‌های گوشت مرغ، گروه B از ایزوله‌های مدفوع انسانی، گروه D از ایزوله‌های گوشت چرخ کرده، گوشت مرغ و مدفوع انسان و گروه E از ایزوله‌های گوشت چرخ کرده تشکیل شدند. شناسایی بیشتر این سویه‌ها به وسیله آنالیز انگشت نگاری (GTG) 5 بر اساس توالی‌های تکرار شده، تمایز بیشتری میان انواع مختلف سویه‌ها نشان نداد. به عنوان نتیجه‌گیری می‌توان گفت که RAPD-PCR روشی مناسب به منظور یافتن منشا آلودگی در زمینه بهداشت و صنعت مواد غذایی می‌باشد.
