اثر سیستئین و ال-کارنیتین بر یک پارچگی DNA اسپرم پس از خروج از انجماد منی فریز شده بز نر

چکیده

پیشینه: افزودن آنتی اکسیدان‌ها به رقیق کننده‌های مایع منی، به منظور محدود کردن تغییرات اکسیداتیو ناشی از انجماد امری ضروری است. هدف: این مطالعه با هدف بررسی اثرات سیستئین و ال-کارنیتین (LC) بر معیارهای اسپرم پس از خروج از انجماد، انجام شد. روش کار: نمونه‌های منی یک بار در هفته از پنج بز نر بالغ نژاد Zaraibi به مدت 10 هفته متوالی جمع آوری شد. نمونه‌های منی تازه از نظر ویژگی‌های اولیه منی مورد ارزیابی قرار گرفتند و نمونه‌های پذیرفته شده با هم ترکیب و به هفت قسمت تقسیم شدند: کنترل (رقیق کننده زرده تخم مرغ تریس)، سیستئین (5/2، 5 و 5/7 میلی مولار) و LC (5/2، 5 و 10 میلی مولار). مقسم‌ها، رقیق و تحت روش حفاظت سرمایی (cryopreservation) قرار گرفتند. پس از خروج از انجماد، ویژگی‌های اسپرم بر اساس تحرک، زنده مانی، یک پارچگی آکروزوم و غشای پلاسمایی (آزمون تورم هیپواسمتیک، HOST)، ظرفیت آنتی‌اکسیدان تام، سطح مالون‌دی‌آلدهید (MDA) و سنجش دنباله‌دار            (comet assay) DNA مورد ارزیابی قرار گرفت. نتایج: افزودن 10 میلی ‌مولار سیستئین و 5/7 میلی ‌مولار LC به طور معنی‌داری سبب افزایش تحرک، زنده مانی، HOST و یک پارچگی آکروزوم پس از خروج از انجماد اسپرم‌ها شد. روش سنجش دنباله‌دار کاهش معنی‌داری را در آسیب DNA با بهترین نتایج در 5/2 میلی مولار سیستئین و 5/7 میلی مولار LC نشان داد. علاوه بر این 10 میلی مولار سیستئین و 5/7 میلی مولار LC کاهش معنی‌داری را در مقدار MDA نشان دادند. سیستئین با زنده‌مانی اسپرم‌ها، یک پارچگی آکروزوم و سنجش دنباله‌دار همبستگی مثبت اما با سطوح MDA و طول دم همبستگی منفی داشت. همبستگی مستقیم بین LC و حرکت پیش رونده، زنده مانی HOST و TCA مشاهده شد در صورتی که همبستگی منفی با LC و MDA، دنباله‌دار، tail DNA، tail moment و olive tail moment وجود داشت. نتیجه‌گیری: افزودن سیستئین و LC به اسپرم منجمد بز سبب بهبود ویژگی‌های اسپرم و کاهش آسیب‌های DNA شد که می‌توان این اثرات مثبت را به اثر بخشی آن‌ها در سرکوب تشکیل MDA نسبت داد.
