حرکت و بازیابی اسپرماتوزوآی آلپاکا (ویکوگنا پاکوس) پس از سانتریفیوژ کردن در یک محلول دارای شیب چگالی

چکیده

پیشینه: یکی از فاکتورهایی که فرآوری موفقیت آمیز منی آلپاکا (ویکوگنا پاکوس) را محدود می‌کند، ویسکوزیته پلاسمای منی است. ماهیت ویسکوز منی انزال شده از آلپاکا مانع انجماد اسپرم و همچنین تلقیح مصنوعی (AI) در شرایط مزرعه می‌شود. هدف: هدف از انجام این تحقیق ارزیابی بازیابی، حرکت و یکپارچگی غشای پلاسمایی اسپرماتوزوآی آلپاکا پس از سانتریفیوژ شدن در یکی از دو محلول مختلف، در یکی از 3 ترکیب متفاوت سرعت و زمان بود. روش کار: در مجموع 24 مایع انزال شده از 7 آلپاکای نر نژاد هواکایا و به ظاهر سالم از نظر تولید مثل، با استفاده از یک واژن مصنوعی (AV) اصلاح شده، پس از آموزش حیوانات برای جمع‌آوری مایع منی، جمع‌آوری شد. جهت انجام این مطالعه، از یک آرایش تیماری 2 در 3 فاکتوریل استفاده شد. منی‌های جمع‌آوری شده جهت سانتریفیوژ شدن در یکی از دو محلول (تریس اکستندر یا محلول دارای شیب چگالی PureSperm®80) در یکی از 3 ترکیب متفاوت سرعت و زمان (× g 492 به مدت 15 دقیقه، × g 1968 به مدت 10 دقیقه، یا × g 4448 به مدت 7 دقیقه) به قسمت‌های مختلف تقسیم شدند. آزمایش 8 بار تکرار شد. نتایج: تجزیه و تحلیل داده‌ها نشان داد که سانتریفیوژ در × g 4448 به مدت 7 دقیقه در محلول PureSperm®80، میزان بازیابی بالایی از اسپرم‌ها با بیشترین تحرک و یکپارچگی عملکردی غشای پلاسمایی را پس از سانتریفیوژ، فراهم می‌کند. نتیجه‌‌گیری: نتایج نشان می‌دهد که استفاده از این روش (سانتریفیوژ در × g 4448 به مدت 7 دقیقه در محلول PureSperm®80) در فرآوری اولیه منی آلپاکا ممکن است که سبب بهبود قابلیت استفاده از اسپرم برای AI و نیز سایر زیست فناوری‌های تولید مثلی کمکی، در این گونه شود.
