خصوصیات مولکولی ژن هماگلوتینین-نوروآمینیداز در بین ویروس‌های حاد نیوکاسل در ایران

چکیده

سویه‌های بیماری‌زای ویروس بیماری نیوکاسل (vNDV) در سراسر دنیا عامل بروز خسارات چشمگیر اقتصادی در صنعت پرورش طیور محسوب می‌شوند. با وجود بومی بودن بیماری نیوکاسل (ND) و وقوع واگیری‌های راجعه، ماهیت و ویژگی‌های ژنتیکی سویه‌های در حال چرخش NDV در ایران به ‌شکل گسترده ناشناخته است. هدف: در مطالعه حاضر 13 جدایه NDV که از موارد رخدادهای بیماری در مناطق مختلف ایران طی سال‌های 1377 تا 1378 جداسازی شده بودند، به وسیله تعیین توالی کامل و آنالیز فیلوژنیک ناحیه کد شونده ژن هماگلوتینین-نوروآمینیداز (HN) تعیین هویت شدند. روش کار: همه جدایه‌ها بر اساس شاخص‌های بیماری‌زایی درون‌تن و توالی آمینو اسیدی (aa) پروتئین همجوشی (F) در محل شکاف (FPCS) مورد تجزیه و تحلیل قرار گرفتند. نتایج: آنالیز فیلوژنیک بر اساس توالی کد شونده ژن HN، ارتباط بسیار نزدیک این ویروس‌ها با ویروس‌های متعلق به ژنوتیپ XIII و تحت ژنوتیپ XIIIa را نشان داد. آنالیز توالی نوکلئوتیدی (nt) ژن HN نشان داد که در تمامی جدایه‌ها پروتئینی به طول 571 اسید آمینه کُد می‌شود و طویل شدن کربوکسیل پایانی در هیچکدام از توالی‌های اسید آمینه‌های پروتئین HN وجود ندارد. آنالیز توالی، جابه‌جایی‌های متعدد اسیدهای آمینه را در جایگاه‌های پادگنی یا اپیتوپ‌های خنثی کننده پروتئین بر روی گلیگوپروتئین HN جدایه‌های مورد مطالعه در مقایسه با سویه‌های متداول واکسینال نمایان ساخت. نتیجه‌گیری: در این مطالعه، خصوصیات مولکولی جدایه‌های بیماری‌زای vNDV مزارع طیور تجاری در ایران به وسیله تعیین توالی کامل و آنالیز ژن HN صورت گرفت. جداسازی و مطالعه خصوصیات مولکولی تعداد بیشتری جدایه NDV از دیگر نواحی کشور و کشورهای همسایه در راستای تعیین ماهیت و منشاء ویروس‌های بومی می‌تواند مفید باشد.
