بررسی ارتباط پلی‌مرفیسم ژن 2 و 6 گیرنده Toll-like با ورم پستان بالینی و صفات تولیدی در گاو هلشتاین

چکیده

ورم پستان یک بیماری هزینه بر گاو شیری است که سبب کاهش میزان تولید و کیفیت شیر در گاوهای مبتلا می‌شود. ژن‌های گیرنده   toll-like (TLR) نقش مهمی در پاسخ میزبان به انواع مختلف ارگانیسم‌ها شامل انواع القا کننده ورم پستان دارند. در مطالعه حاضر، پلی‌مرفیسم ژن‌های 2، 4، 6 و 9TLR در گاو هلشتاین و ارتباط آن‌ها با ورم پستان بالینی (CM)، امتیاز سلول‌های سوماتیک شیر (SCS) و صفات تولیدی شیر مورد بررسی قرار گرفت. از گله بزرگ تجاری هلشتاین؛ 38 عدد نمونه خون جمع آوری شد. 19 مورد، از گاوهای بدون سابقه قبلی ورم پستان (غیر حساس) و 19 مورد، از گاو‌های هلشتاین با حداقل 3 مورد قبلی ابتلا به ورم پستان (حساس). تعیین ساختار ژنتیکی 4 ژن TLRs با استفاده از واکنش زنجیره‌ای پلیمراز-چند شکلی طولی قطعات محدود الاثر (PCR-RFLPs) و آنزیم‌های Rsa1، MSP1، Hha1، HaeIII و Taq1 انجام شد. هفت پلی‌مرفیسم‌های تک نوکلئوتیدی (SNPs) غیر متشابه جدید در ژن‌های 2 ،4 و 6TLR در حیوانات حساس یافت شد. ارتباط بین پلی‌مرفیسم ژن Taq1-TLR2 با ورم پستان بالینی، چربی شیر و حداکثر میزان تولید شناسایی شد. علاوه بر این ارتباط بین Taq1-TLR6 با پیک تولید (PY) و تداوم شیردهی نیز نشان داده شد. جهش‌ها در TLRsها که به کرات در گاوهای حساس گزارش شده است، نشانگرهای ژنتیکی بالقوه‌ای را برای شناسایی (MAS) موارد مقاوم به ورم پستان در گاوهای شیری فراهم می‌کند.
