Background: Canine parvovirus, an important and highly pathogenic agent for puppies, is distributed worldwide. Despite the various vaccines being available, it is not easily prevented. Immunoglobulin therapy has been attempted as a supportive approach to cure affected puppies. Aims: In order to achieve a homologous immunoglobulin G (IgG) for the treatment of the fatal parvovirus, the purification of anti-parvovirus IgG from immunized dogs was assayed. Methods: IgG purification was conducted on the obtained immune sera using two-stage dialysis, in which various concentrations of ammonium sulfate with several incubations and centrifugations were applied in multiple steps. Results: Using SDS-PAGE electrophoresis, the correct band sizes of the purified IgGs were observed and its concentration was assessed to be >22 mg/ml. Also, using quantitative ELISA, the specific anti-parvovirus IgG titer of the purified dog IgG was calculated to be considerably above the protective threshold titer. To do so, 13 parvoviral diarrheic puppies were randomly categorized into 3 treatment groups for 5 consecutive days. Administration of 4, 8 and 16 mg/kg bw/d of purified IgG to these puppies significantly improved all clinical symptoms including diarrhea, dehydration, appetite, vomiting and white blood cell (WBC) in all affected puppies in dose-dependent manner, comparing with those in the placebo-controlled group (n=8) that experienced 2 deaths, too. Conclusion: This homologous anti-parvovirus IgG, which was purified through a novel, simple and inexpensive technology is expected to pass further clinical trials. Also, this IgG has the potential as a commercial product to reduce the burden of parvovirus infection in puppies.

Background: Aging is associated with various physiological changes that lead to oxidative stress and the production of reactive oxygen species (ROS), negatively affecting sperm structure and function. Aims: This study aimed to investigate the impact of incorporating rosemary essential oil and selenium yeast into the diet on sperm quality parameters, antioxidant enzyme activity, and lipid peroxidation rates of semen in aged broiler breeder roosters. Methods: Thirty-six 57-week-old Ross 308 roosters were randomly assigned in a 3×2 factorial arrangement, in six treatments with six replications. The treatments included three levels of rosemary essential oil (0, 100, and 200 mg/kg) and two levels of selenium yeast (0 and 0.3 mg/kg) for 7 weeks. Sperm parameters, antioxidant enzyme activities, and lipid peroxidation levels in seminal plasma were evaluated. Results: The results showed significant effects of rosemary essential oil and selenium yeast on sperm parameters. 100 mg rosemary essential oil plus 0.3 mg selenium yeast/kg of diet showed the most significant improvement in sperm quality (P<0.05). However, there was no significant difference in sperm morphological abnormalities among the treatments. Additionally, seminal plasma antioxidant enzyme activities, including glutathione peroxidase (GPx), superoxide dismutase (SOD), catalase (CAT), and total antioxidant capacity (TAC) significantly increased and malondialdehyde (MDA) decreased, particularly in the treatment containing 100 mg rosemary essential oil plus 0.3 mg selenium yeast/kg diet (P<0.05). Conclusion: Overall, 100 mg/kg rosemary essential oil combined with 0.3 mg/kg selenium yeast in the diet is recommended for aged broiler breeder roosters to enhance their reproductive function.

Background: High stocking density is one of the main factors that impair the performance and welfare of broilers. Aims: This study aimed to investigate the effects of Bacillus and Lactobacillus probiotics, either alone or in combination, on meat quality, peroxide value of fat tissue, leg health, and litter quality in broilers under high stocking density. Methods: Two hundred one-day-old Cobb 500 chicks were divided randomly into five treatments (4 replicates and ten birds per replicate): Control (normal stocking density, no probiotic), HD (high stocking density, no probiotic), HDpBacil (high stocking density, Bacillus-based probiotic), HDpLAB (high stocking density, Lactobacillus-based probiotic), HDpMix (high stocking density, combined probiotic containing Bacillus and Lactobacillus). Litter quality was assessed on days 21 and 42. Footpad dermatitis was also scored on days 14, 28, and 42. Gait score, peroxide value in fat tissue, and breast and thigh meat quality were measured on day 42. Results: The HD group had significantly the highest footpad dermatitis and gait scores and the lowest litter quality levels (P<0.05). The combined probiotic improved leg health and litter quality indices (P<0.05) and reduced the water holding capacity (WHC) and peroxide value (P<0.05). While, the highest peroxide value of abdominal fat and WHC of thigh meat were in the HD group (P<0.05). Conclusion: The combination of probiotics (Bacillus and Lactobacillus) can mitigate the adverse effects of high stocking density in broilers. It can be a beneficial feed additive in broilers against highly stressful conditions.

Background: Application of dietary phytobiotics against bacterial pathogens in shrimp farming is considered a promising alternative to antibiotics. Aims: The present study investigated the impact of tamarind pulp-powder (TPP) supplemented diets on growth, survival, immune responses, resistance to Vibrio harveyi, and immune gene expression in Litopenaeus vannamei. Methods: A 49-day feeding trial was conducted using L. vannamei (mean weight 5.60±0.45 g) fed with TPP-supplemented diets containing 0 (T0), 1 (T1), 5 (T2), and 10 (T3) g/kg diets at 3% body weight. Growth performance viz., net weight gain (NWG) and feed conversion ratio (FCR) were recorded on weekly basis and immune responses viz., serum protein (SP), Prophenoloxidase (PPO), and lysozyme (LZ) activities were analyzed at the end. A challenged test conducted for 96 h with V. harveyi (1×105 CFU/ml) to assess mortality. After the challenge test, enzymatic activities and immune genes viz., toll-like receptor and lysozyme were analyzed. Results: Shrimp fed with the T2 diet (5 g/kg TPP) showed significantly (P< 0.05) higher NWG, improved FCR, and survival compared to other groups. Total hemocyte count, SP, PPO, and LZ activities were significantly higher in T2 shrimp. Reduced levels of enzymatic activities and upregulated expression of immune-related genes were also observed in T2. During the challenge, T2-fed shrimp achieved the highest survival rate (86%), confirming enhanced resistance to V. harveyi. Conclusion: A dietary inclusion level of 5 g/kg TPP was found optimal for improving growth, immunity, and resistance against vibriosis in L. vannamei, highlighting its potential as a phytobiotic alternative to antibiotics in shrimp aquaculture.

Background: Our previous study demonstrated that exopolysaccharides (EPS) from Lactobacillus plantarum (L. plantarum) act as an effective adjuvant. They can enhance both the immune response elicited by the oprH recombinant subunit vaccine against Pseudomonas aeruginosa (P. aeruginosa) and the protective efficacy in experimental animals. However, it is not clear whether oral administration of L. plantarum EPS can promote immune function in experimental animals. Aims: The present study aimed to explore the immunoregulatory function of L. plantarum EPS in mice. Methods: EPS was extracted from L. plantarum. EPS was then orally administered to the mice at doses of 20, 40, and 60 mg/kg for 20 consecutive days. After 10 and 20 days, the immune organ index, delayed-type hypersensitivity (DTH), phagocytic ability of peritoneal macrophages, serum hemolysin, and splenic lymphocyte proliferation of mice were measured. The concentrations of IgG1, IgG2a, IFN-γ, IL-2, IL-4, and IL-10 in serum were measured using ELISA. The percentage of CD4+ and CD8+ T lymphocytes subpopulations and the expression of transcription factors T-bet and GATA-3 in the mouse spleen cells were measured by flow cytometry and qRT-PCR, respectively. Results: L. plantarum EPS could induce high levels of immune organ index, DTH reaction, phagocytic ability of peritoneal macrophages, serum hemolysin, splenic lymphocyte proliferation, serum IgG1, IgG2a, Th1-type cytokines IL-2, IFN-γ, Th2-type cytokines IL-4, IL-10, CD4+ T lymphocyte, CD8+ T lymphocyte, T cell transcription factors T-bet and GATA-3, and has a certain dose-dependency. Conclusion: L. plantarum EPS can concurrently upregulate both Th1 and Th2 immune responses in mice, with this effect being more pronounced, particularly at the high dose. Our study provides a theoretical basis for research on L. plantarum EPS in veterinary.

Background: Working canines play a very important role in assisting forces by uncovering evidences that might be missed by the human eye. To date, there is no published data on normal ophthalmological parameters for working dogs. Aims: This study aims to document normal reference ranges of ophthalmological parameters and blood pressure (BP). Additionally, the study compared the two methods of intraocular pressure (IOP) measurement and A-scan biometry, and assess correlations between IOP to both systolic blood pressure and axial globe length. Methods: The study was conducted on 18 dogs of Sashastra Seema Bal (SSB), Alwar (Rajasthan), aged 3.5-6 years, which were divided into three breed-specific groups, i.e., group 1 (Belgian Shepherd), group 2 (German Shepherd) and group 3 (Labrador Retriever) with six animals each. Schirmer’s tear test strips were used to perform the Schirmer’s tear test (STT). To measure intraocular pressure (IOP), rebound tonometry (Tonovet) and indentation tonometry (Schiotz) were used. A-scan ultrasonography was performed by contact and immersion method. Fundoscopic images were recorded using Fundus-On-Phone equipment. Results: STT values revealed non-significant differences among groups, sexes, and eyes. The cumulative mean reflected a higher reading by the rebound tonometer than the indentation tonometer. In all the three groups, a significantly longer anterior chamber depth and axial globe length were observed in the immersion method. Axial globe length was longest in the Labrador breed, followed by German Shepherd and Belgian Shepherd. Systolic and diastolic pressure were significantly different among groups. Conclusion: These breed-specific various ophthalmic parameters of working canines will be useful in the clinical diagnosis of various ophthalmic affections.

Background: Essential oils and organic acidifiers are used as natural antimicrobial substitutes for chemical additives in poultry feed. Pathogens such as Salmonella and Escherichia coli pose significant risks. While essential oils possess strong antibacterial and antifungal properties, commercial acidifiers (blends of formic, propionic, and lactic acids) are also commonly applied to reduce microbial contamination in feed. However, comparative data on their efficacy against key poultry pathogens remain limited. Aims: ‏The present study investigates the antibacterial and antifungal effects of selected commonly used commercial acidifiers in poultry feed and also compares them with antibacterial effects of essential oils. Methods: Antibacterial effects against Salmonella typhimurium and E. coli were evaluated for both five commercial acidifiers and essential oils (cinnamon, thyme, clove). Antifungal effects against Fusarium oxysporum were evaluated for acidifiers only. Microplate and agar-well diffusion methods were used. Acidifiers were tested at 1×, 2×, and 5× the manufacturer’s recommended dose; essential oils at 2% and 5% (v/v). Sampling time points for feed pH and microbial counts were days 0, 7, and 21. All tests were performed in triplicate and repeated twice. The impact of acidifiers on feed pH was also evaluated. Results: Acidifiers exhibited negligible inhibitory effects against target pathogens even at concentrations five times higher than recommended levels. In contrast, essential oils demonstrated significant inhibitory effects. Furthermore, the addition of acidifiers did not affect the pH of the feed. Conclusions: The study underscores the potent antibacterial properties of cinnamon, thyme, and clove essential oils against common poultry pathogens. Comparatively, commercial acidifiers showed limited efficacy. Incorporating essential oils into poultry feed can enhance animal health and safety as effective substitutes for chemical additives. Further research is needed to optimize their use and maximize their antimicrobial benefits.

Background: Footrot is a highly contagious disease of the feet in sheep and goats. Aims: The study was conducted to determine the serogroup distribution of Dichelobacter nodosus in sheep affected by footrot on organized farms and flocks reared by farmers across four districts of the Kashmir Valley. Methods: Sheep from randomly selected organized farms and flocks raised by farmers and the Bakerwal tribes were screened for footrot lesions based on clinical signs, particularly lameness. Two hundred sixty swab samples were collected from sheep exhibiting footrot lesions, and D. nodosus infection was confirmed using 16S rRNA-specific PCR. For serogroup subtyping, multiplex PCR was performed. Furthermore, we utilized the pET28a expression vector to clone and express the outer membrane fimbrial protein antigen, which was confirmed using SDS-PAGE and Western blot analysis. Results: D. nodosus was detected in 149 samples by 16S rRNA-specific PCR. Serogroups B and E were detected in 85.9% and 10.74% of positive samples, respectively, while both serogroups were detected in 3.36% of the samples. The outer membrane domain containing the major antigenic epitopes of the pilQ gene of D. nodosus was successfully cloned into the pET28a expression vector. Conclusion: The prevalence of ovine footrot was found to be low during the study period. Serogroup B of D. nodosus was found to be predominant, while serogroup E was detected exclusively in some isolated flocks reared by farmers. The pilQ gene was successfully expressed in Escherichia coli and purified to near homogeneity, making it suitable for further exploitation as a serogroup-independent vaccine candidate against footrot.
