		<Abstract>Botulism is a serious paralytic disease caused by Clostridium botulinum toxin in foods. There are seven recognized serotypes of botulinum neurotoxins among which the principal prevalent types in humans include A, B and E. Infant botulism results from intestinal colonization and toxin production by C. botulinum spores in babies less than 1 year old. Honey is the most important food discriminated as the cause of infant botulism. In this study, a total of 100 honey samples were collected from retail markets in Shiraz,&lt;br /&gt;Iran and analysed for the presence of C. botulinum using a multiplex PCR assay. Three pairs of primers were designed and optimized to identify A, B and E strains in the specimens. Positive samples were further examined to find out whether they carried the toxin gene; toxin products were also checked by mouse bioassay. The results showed that out of 100 honey samples tested, 2% were found positive for type B which carried the neurotoxin B gene confirmed by sequencing. All the injected mice died, whereas no symptoms&lt;br /&gt;were observed in the control groups. The honey collected from Shiraz retail markets was contaminated with the spore of the bacteria and can thus potentially cause infant botulism. Bioassay is described as a standard method; however, molecular based techniques also easily, quickly and reliably detect C. botulinum, its toxins and spores in food stuffs and is strongly advised for use in the food microbial lab.</Abstract>
			<OtherAbstract Language="FA">بوتولیسم یکی از بیماری‌های جدی فلج کننده است که در اثر سم باکتری &lt;em&gt;کلستریدیوم بوتولینوم&lt;/em&gt; در مواد غذایی ایجاد می‌شود. تعداد 7 سروتیپ مشخص از نوروتوکسین بوتولینوم شناسایی گردیده است که در میان آن‌ها اصلی‌ترین تیپ‌های شایع در انسان شامل تیپ‌های A، B و E می‌باشد. بوتولیسم نوزادان در نتیجه تجمع میکروارگانیسم‌ها در روده و تولید سم می‌باشد که در اثر حضور اسپور باکتری در نوزادان زیر یک سال سن ایجاد می‌گردد. عسل یکی از مهمترین غذاهایی است که در ایجاد بوتولیسم نوزادان نقش دارد. در این مطالعه تعداد صد نمونه از مراکز فروش عسل در شیراز جمع آوری گردید و با استفاده از روش واکنش زنجیره‌ای پلیمراز چند تایی مورد ارزیابی قرار گرفت. بدین منظور از سه جفت پرایمر طراحی و بهینه شده برای شناسایی قطعی گونه‌های A، B و E در نمونه‌ها استفاده گردید. نمونه‌های مثبت مورد آزمایش تکمیلی قرار گرفتند که ضمن قطعی نمودن وجود ژن تولید کننده سم، سم باکتری با استفاده از آزمون حیاتی موش آزمایش گردید. نتایج نشان دادند که 2% از نمونه‌ها آلوده به تیپ B باکتری می‌باشند که در نهایت پس از انجام توالی‌یابی مشخص گردید که حامل ژن تولید کننده نوروتوکسین B نیز بوده‌اند. مرگ موش‌ها در گروه‌های آزمایشی در آزمون حیاتی موش قطعی گردید. عسل‌های جمع آوری شده نشان دهنده آلودگی به اسپور باکتری بوده است و بنابراین می‌تواند در ایجاد بوتولیسم نوزادان نقش داشته باشد. هر چند که آزمون حیاتی موش به عنوان یک روش استاندارد توصیف شده است، روش‌های مولکولی، ساده، سریع و مطمئن برای شناسایی تیپ‌های مختلف باکتری یا سموم آن‌ها بوده و قابل توصیه در آزمایشگاه‌های بهداشت مواد غذایی می‌باشد.</OtherAbstract>
