Poultry and poultry products are known as important sources of human campylobacteriosis. In this study, during the autumn months of 2005, a total of 100 samples from poultry carcasses, representing 20 broiler flocks were obtained by rinse test, after the chilling stage of processing. The samples were enriched in Preston broth, followed by streaking on selective media. Then, the suspected colonies were isolated on sheep blood agar and tested for morphology, motility and Gram-staining. Biochemical tests and hippurate hydrolysis activity were also performed. Concurrently, a multiplex PCR assay (m-PCR) with two sets of primers was employed for identification of Campylobacter  genus and Campylobacter coli. The m-PCR assay was applied on bacterial cultures harvested from selective media plates. By conventional culture method, including hippurate hydrolysis test from suspected colonies, 76% of samples were positive for Campylobacter spp. and 2% for C. coli. In m-PCR assay 28% of the harvested cultures, were positive for Campylobacter genus but C. coli  were not detected in any of the samples. According to this preliminary study, it seems that the contamination rate of poultry carcasses with other species of Campylobacter genus is higher than contamination with C. coli.  }, keywords = {Campylobacter coli,Campylobacter spp,m-PCR,Poultry carcass}, title_fa = {جداسازی و شناسایی جنس کمپیلو باکتر و گونه کمپیلو باکتر کولای با استفاده از روش کشت مرسوم و روش multiplex-PCR از لاشه طیور کشتار شده در مشهد، ایران}, abstract_fa = {گونه‌های مختلف جنس کمپیلو باکتر که عامل بسیاری از موارد گاستروآنتریت در انسان می‌باشند، از نظر بهداشت عمومی از اهمیت ویژه‌ای برخوردار می‌باشند. گوشت طیور به عنوان منشا مهم کمپیلوباکتریوزیس در انسان شناخته شده است. در این مطالعه تعداد یکصد لاشه طیور متعلق به 20 گله طیور گوشتی با استفاده از روش تست شستشو (rinse test) مورد نمونه‌برداری قرار گرفت. پس از کشت در محیط غنی کننده آبگوشت پرستون و کشت در آگار انتخابی از هر یک از کلنی‌های مشکوک، در محیط آگار خون‌دار کشت داده شد. کلنی‌های جدا شده از نظر مورفولوژی باکتری، رنگ‌آمیزی گرم، حرکت، تست‌های بیوشیمیایی و هیدرولیز هیپورات مورد آزمایش قرار گرفتند. همچنین از کلنی‌های مشکوک تست واکنش زنجیره‌ای چند گانه‌ای (multiplex-PCR) با استفاده از پرایمرهای اختصاصی صورت گرفت که مشخص کننده جنس کمپیلو باکتر و نیز گونه کمپیلو باکتر کولای می‌باشند. در روش کشت مرسوم، 76% و در روش m-PCR، 28% از نمونه‌ها آلوده به جنس کمپلو باکتر تشخیص داده شد. در کشت مرسوم 2% آلودگی به کمپیلو باکتر کولای تشخیص داده شد ولی در تست m-PCR آلودگی به کمپیلو باکتر کولای مشخص نگردید. بر اساس این مطالعه مقدماتی به نظر می‌رسد آلودگی لاشه طیور به سایر گونه‌های جنس کمپیلو باکتر بیشتر از گونه کمپیلو باکتر کولای باشد.}, keywords_fa = {کمپیلو باکتر کولای,جنس کمپیلو باکتر,روش m-PCR,لاشه طیور}, url = {https://ijvr.shirazu.ac.ir/article_536.html}, eprint = {https://ijvr.shirazu.ac.ir/article_536_9dfb7d19c76381e673a5245bbc751536.pdf} }
