The chicks were divided into four groups [group I (NSS), group II (LPS), group III (Vit. D3 + NSS), and group IV (Vit. D3 + LPS)] containing eight chicks in each group, treated accordingly for 21 days. Birds were kept under close observation for apparent clinical signs and symptoms. Clinically, vitamin D3 treated chicks were dull, off feed, showed polydipsia, polyuria, watery faeces, rigidity of limbs, severe dehydration, weakness and significant progressive emaciation. Grossly, the bones were soft whereas most organs revealed congestion and hemorrhages in visceral organs. Histopathologically, renal tubular epithelium showed coagulative necrosis and metastatic calcification. The lung parenchyma and bronchi showed hemorrhages, congestion with diffuse heterophilic cell infiltration in inter-alveolar septa and infiltration of heterophils in alveoli along with proteinacious fluid in LPS treated chicks of treatment group, suggesting immunomodulatory action of vitamin D3. Vitamin D3 treated chicks showed strong expression of Calbindin D28k in intestine and kidney but weak expression in lung, which can be linked to nephrocalcinosis seen in kidney and from its prospective role in cellular calcium homeostasis.}, keywords = {Broiler chick,Calbindin D28K,Cholecalciferol,Immunomodulation,LPS}, title_fa = {هیپرویتامینوز D3 در جوجه‌های گوشتی: روش هیستوپاتولوژی، ایمونومدولاتوری و ایمونوهیستوشیمی}, abstract_fa = {مطالعه حاضر به منظور بررسی اثرات سمی دوزهای بالاتر (1,00,000 IU یعنی 5/2 میلی‌گرم/کیلوگرم وزن بدن) ویتامین D3، همزمان با اندوتوکسین‌های باکتریایی (لیپوپلی ساکارید: LPS) انجام گرفت تا پتانسیل تعدیل کنندگی سیستم ایمنی یا ایمونومدولاتوری ویتامین D3 در جوجه‌های گوشتی IBL-80 مورد مطالعه قرار گیرد. جوجه‌ها به 4 گروه [گروه I (NSS)، گروه II (LPS)، گروه III (Vit. D3 + NSS) و گروه IV (Vit. D3 + LPS)] شامل 8 جوجه در هر گروه تقسیم شدند، بر این اساس به مدت 21 روز درمان شدند. پرندگان تحت مشاهده نزدیک از نظر علایم و نشانه‌های مشخص بیماری نگهداری شدند. از لحاظ بالینی، جوجه‌های درمان شده با ویتامین D3 بی‌حال و نامتمایل به غذا بودند و پر نوشی، پر ادراری، مدفوع آبکی، سفتی اندام‌های حرکتی، کم آبی شدید، ضعف و لاغری پیشرونده قابل توجهی را نشان دادند. از نظر ظاهری، استخوان‌ها نرم بودند در حالی که اغلب ارگان‌ها پر خونی و خونریزی در اندام‌های احشایی را تایید کردند. از نظر هیستوپاتولوژی، اپی تلیوم توبول‌های کلیوی نکروز انعقادی و کلسیفیکاسیون متاستاتیک را نشان دادند. پارانشیم ریه و برونش‌ها خونریزی، پر خونی همراه با نفوذ منتشر سلول‌های هتروفیل در دیواره بین آلوئولی و نفوذ هتروفیل‌ها در آلوئول‌ها همراه با مایع پروتئینی در جوجه‌های درمان شده با LPS گروه درمان را نشان دادند که فعالیت ایمونومدولاتوری ویتامین D3 را نشان می‌دهد. جوجه‌های درمان شده با ویتامین D3 بیان قوی کالبیندین D28k در روده و کلیه اما بیان ضعیف آن در ریه را نشان دادند که می‌تواند مربوط به نفروکلسینوز مشاهده شده در کلیه و به واسطه نقش آن در همئوستاز کلسیم سلولی باشد.}, keywords_fa = {جوجه گوشتی,کالبیندین D28K,کوله کلسیفرول,ایمونومدولاسیون,LPS}, url = {https://ijvr.shirazu.ac.ir/article_4218.html}, eprint = {https://ijvr.shirazu.ac.ir/article_4218_8f6cd677af94e1fedc248508dcec6654.pdf} }
