The hypothesis tested in this investigation is; arsenic exposure causes neural tube defects (NTDs) andthese defects of the central nervous system are more likely related to folate deficiency during fetal life. In this study, sodium arsenate was administered via intraperitoneal route at a rate of 40 mg/kg body weight on the 6th day of gestation to every individual of 20 mated Albino mice. On the 8th (E8), 10th (E10), 16th (E16) and 19th (E19) days of the gestation, the pregnant mice in control and experimental groups were sacrificed by cervical dislocation. All embryos belonging to (E16) and (E19) were examined for external morphological neural tube defects. Histological staining techniques were haematoxylin and eosin and theimmunofluorescence staining was also implemented. It was observed that, the intraperitoneal injection ofsodium arsenate caused a number of morphological neural tube defects including; open fourth ventricle,exencephally, myelomeningocele and anencephaly. Difference in control and experimental specimens wassignificant (P<0.001) on the (E16) group. The histomorphologic changes of neural tube were significant in allof the experimental groups in comparison to the controls. Immunofluorescence study revealed reduced folate carrier (RFC-1) protein reduction in neural tissue, and these results demonstrate that the association between prenatal exposure to inorganic arsenic and NTDs is more likely related to a defect in reduced folate carrier protein.}, keywords = {Arsenic,Neural tube,Mouse embryo,RFC-1}, title_fa = {تاثیر آرسنیک بر لوله عصبی جنینی موش و ارتباط آن با پروتئین RFC-1}, abstract_fa = {آرسنیک، از مواد سمی محیط اطراف است که به صورت غیر آلی در آب مصرفی وجود دارد. فرضیه مورد نظر در این تحقیق این است که آرسنیک باعث بروز نقایص لوله عصبی می‌شود که احتمالا با کاهش دریافت فولیت ارتباط دارد. در این مطالعه سدیم آرسنیک از طریق داخل صفاقی با دوز mg/kg40 وزن بدن در روز ششم حاملگی به 20 موش باردار تزریق شد. جنین‌های مربوط به گروه‌های شاهد و آزمایشی در روزهای 19، 16، 10 و 8 جنینی از رحم خارج شدند. تمامی جنین‌های شانزده و نوزده روزه تحت مطالعه مورفولوژیکی ناحیه لوله عصبی قرار گرفتند و مطالعات هیستومورفولوژیک از طریق رنگ‌آمیزی‌های (H&E) و (I.F) ایمونونوفلورسانسی انجام شد. تزریق 40 میلی‌گرم بر کیلوگرم سدیم آرسنیک، منجر به بروز نقایص مورفولوژیکی شامل باز بودن بطن چهارم مغزی، اگزنسفالی، میلومننگوسل و آنانسفالی شد. مقایسه دو گروه شاهد و آزمایشی نشان دهنده وجود ارتباط معنی‌دار در روز 16 جنینی بود، همینطور مطالعه هیستومورفولوژیک لوله عصبی وجود اختلاف معنی‌دار بین دو گروه را در روزهای 19، 16، 10 و 8 را ثابت کرد. بررسی‌های میکروسکوپیک با استفاده از رنگ‌آمیزی ایمونوفلورسانس، نشان داد که میزان پروتئین 1-RFC در بافت عصبی کاهش یافته و احتمالا نقایص ایجاد شده در لوله عصبی توسط آرسنیک در ارتباط با کاهش بروز این پروتئین باشد.  }, keywords_fa = {آرسنیک,لوله عصبی,جنین موش,RFC-1}, url = {https://ijvr.shirazu.ac.ir/article_367.html}, eprint = {https://ijvr.shirazu.ac.ir/article_367_267e930c125cf6b38b64361138abed28.pdf} }
