Ejaculates were collected from four rams. In each session, second ejaculates (n=4) were collected in a tube containing 1 mL coating buffers which were prepared by 10, 15 or 20% egg yolk plus Tris-fructose. Samples were pooled, centrifuged and the supernatant removed in the laboratory. The pellets were diluted with Tris-fructose containing egg yolk which was equal to its concentration in coating. Toperform four replicates for each sample, diluted sample was split into four parts then each part was split into two fractions. One of them (cold-shock) was suddenly put on ice water and the other was chilled at 0.25°C/min until 5°C. Aliquots were kept at 5°C for 36 h and sperm motility, viability and functional membrane integrity were determined at 0, 12, 24 and 36 h. The results showed that functional membrane integrity was highest in 20% yolk egg under gradual cooling at 0, 24 and 36 h (85.7, 75.9 and 71.7%,respectively; P<0.05). In the presence of 20% egg yolk, sperm viability was highest under cold-shock (74.8%) and gradual cooling (78.5%) and sperm motility was lower in cold-shock than gradual cooling at all storage times (76.9, 60.7, 36.2 and 27.7% vs. 85.4, 73.1, 54.6 and 40.8%, respectively; P<0.05). Therefore, 20% egg yolk can improve coated sperm longevity in normal gradual cooling; but it does not prevent the destructive effect of cold-shock from taking place.}, keywords = {Coated spermatozoa,Cold-shock,Egg yolk,Ram}, title_fa = {اثر مقادیر مختلف زرده تخم مرغ بر پوشش‌دار کردن اسپرم قوچ و ذخیره سازی در°C 5}, abstract_fa = {این آزمایش جهت ارزیابی اثر سطوح مختلف زرده تخم مرغ برای پوشش‌دار کردن و ذخیره سازی اسپرم انجام شد. انزال‌ها از چهار قوچ جمع‌آوری شد. در هر نوبت، دو انزال از هر قوچ گرفته شد و انزال دوم (12=n) درون لوله حاوی 1 میلی‌لیتر بافر پوشش دهنده که شامل 10، 15 یا 20% زرده تخم مرغ به همراه تریس-فروکتوز بود جمع‌آوری شد. نمونه‌ها تجمیع شدند و پس از سانتریفوژ محلول رویی حذف شد. رسوب‌ها با محلول تریس-فروکتوز حاوی زرده تخم مرغ مساوی با محلول پوشش دهنده رقیق شدند. برای انجام چهار تکرار برای هر نمونه‌گیری، نمونه‌های رقیق شده به چهار بخش تقسیم شد و سپس هر بخش به دو قسمت تقسیم شد. یک قسمت به طور ناگهانی روی آب یخ قرار داده شد (شوک سرمایی) و قسمت دیگری با سرعت °C25/0 در هر دقیقه تا °C5 سرد شد. نمونه‌ها در دمای °C5 به ‌مدت 36 ساعت نگهداری شدند و تحرک، زنده‌مانی و سلامت غشای پلاسمایی آن‌ها در زمان‌های صفر، 12، 24 و 36 ساعت ارزیابی شد. نتایج نشان داد که بالاترین درصد سلامت غشای پلاسمایی با 20% زرده تخم مرغ تحت شرایط سرد کردن تدریجی در زمان‌های صفر، 24 و 36 ساعت به دست آمد (7/85%، 9/75% و 7/71%، P<0.05). در حضور 20% زرده تخم مرغ، بالاترین درصد زنده‌مانی اسپرم تحت شرایط شوک سرمایی (8/74%) و سرد کردن تدریجی (5/78%) به دست آمد و تحرک اسپرم در شرایط شوک سرمایی نسبت به سرد کردن تدریجی در تمام زمان‌های ذخیره‌سازی (به ترتیب 9/76%، 7/60%، 2/36% و 7/27% در مقابل 4/85%، 1/73%، 6/54% و 8/40%، P<0.05). پایین‌تر بود. بنابراین، 20% زرده تخم مرغ می‌تواند سبب بهبود ماندگاری اسپرم را در شرایط سرد کردن تدریجی شود، اما مانع از اثرات مخرب شوک سرمایی نمی‌شود.}, keywords_fa = {اسپرم پوشش‌دار شده,شوک سرما,زرده تخم مرغ,قوچ}, url = {https://ijvr.shirazu.ac.ir/article_2366.html}, eprint = {https://ijvr.shirazu.ac.ir/article_2366_fbde9eb456a0ee0048de2ba4c7651289.pdf} }
