These meats are used in food industry products and/or for direct consumption of consumers. The mitochondrial DNA was amplified as a template in a PCR reaction by use of specific primers related to each species. Specific primers with mitochondrial origin amplified amplicons with the length of 300, 172 and 122 bp in target regions in cattle, sheep and goat, respectively. For determination of the primer sensitivity a set of binary meat mixtures with 0.1, 0.5, 1, 5, 10, 50 and 100% based on weight ratio was tested. The detection limit was found to be 0.1% for all samples. In heat-processing program boiled water for 20 min, 121°C for 20 and 30 min and 127°C for 20 min by autoclaving was used, similar to the conditions that are used in pasteurization, sterilization and also meat processing in industrial factories. The performance of the method was not affected by prolonged heat treatments. DNA میتوکندریایی به عنوان الگو در طی واکنش زنجیره‌ای پلیمراز با استفاده از پرایمرهای اختصاصی مرتبط به هر یک از گونه‌ها مورد تکثیر قرار گرفت. پرایمرهای اختصاصی با منشأ میتوکندریایی به خوبی توانستند نواحی هدف را در گاو به اندازه ٣۰۰، گوسفند ۱٧۲ و بز ۱۲۲ جفت باز تکثیر نمایند. برای تعیین میزان حساسیت پرایمرهای مورد استفاده، از مخلوط‌های دوتایی گوشت به نسبت‌های وزنی ۱/۰، ۵/۰، ۱، ۵، 10، ۵۰ و ۱۰۰ درصد استفاده شد. میزان حساسیت برای تمامی نمونه‌ها 1/0 درصد برآورد شد. در برنامه حرارتی از آب جوش، ۱۲۱ درجه سانتی‌گراد به مدت ۲۰ و ٣۰ دقیقه و ۱۲٧ درجه سانتی‌گراد به مدت ۲۰ دقیقه مشابه شرایط استریلازیسیون، پاستورازیسیون و فرآوری گوشت در کارخانجات صنعتی استفاده شد. تیمار حرارتی طولانی مدت سبب نشد که عملکرد روش به کار گرفته شده تحت تأثیر قرار گیرد. نتایج به دست آمده در این پژوهش، کارآیی استفاده از پرایمرهای اختصاصی گونه‌ای مبتنی بر ژنوم هدف میتوکندریایی را به منظور شناسایی گوشت‌های خالص، مخلوط، خام و حرارت داده شده گاو، گوسفند و بز را با حساسیت و دقت بالا مورد تأیید قرار داد.}, keywords_fa = {آغازگرهای اختصاصی گونه‌ای,گوشت‌های مخلوط,DNA میتوکندریایی,منشأ یابی مواد غذایی}, url = {https://ijvr.shirazu.ac.ir/article_1386.html}, eprint = {https://ijvr.shirazu.ac.ir/article_1386_2051050e86ef4386d445e278db10dfa5.pdf} }
